External accumulation LC-ESI FTICR with automated gain control

An automated gain control (AGC) with external accumulation liquid chromatography (LC)-electrospray ionization (ESI)-Fourier transforms infrared spectroscopy (FTICR) combined with independently controlled internal calibration was developed. Three approaches for data dependent adjustment of ion accumulation in the accumulation quadrupole were evaluated. The efficiency of different AGC approaches in identification of putative peptides was investigated in the course of LC separation of a 0.05 mg/ml solution of four casein proteins digested with trypsin. The adjustment of ion accumulation on the continuous ion current on the conductance limit was found to give rise to the number of putative peptides comparable with that of the conventional LC-ESI-FTIR.
